Description : |
Lubricin/PRG4/MSF/SZP (GenBank NP_005798) is a mucinous glycoprotein which is both a lubricant of artcular cartilage and an anti-adhesive protein. It is expressed by superficial zone articular chondrocytes and synovial fibroblasts, and is also expressed by other organ systems including the liver, brain and heart. The protein has a molecular weight of 206 kDa measured by sedimentation analysis, yet due the post-translational modifications the apparent molecular weight on SDS-PAGE is 280-345 kDa. The variability may be due the 50% (w/w) post-translational modifications which are mainly O-linked β(1-3)Gal-GalNAc and α(2,3)NeuAc -β(1-3)Gal-GalNAc. |

Endotoxin level is less than 0.1 ng per µg (1EU/µg). |
Reconstitution : |
We recommend a quick spin followed by reconstitution in 1x PBS pH 7.2-7.4 to a concentration of 0.25 mg/ml.
This solution can then be diluted into other aqueous buffers and stored at 40C
for 1 week or stored at -200C for future use. |
Biological Activity : |
Synoviocyte binding inhibition1 on Lubricin coated plates at 100 μg/ml, relative to uncoated plates, was 43% for synoviocyte Lubricin and 41% for Lubricin2 from synovial fluid. |
