Description : |
Interleukin 12 (IL-12) is a regulatory protein produced by activated B lymphocytes and macrophages. The biological activities of IL-12 include the stimulation of growth of activated CD4+ and CD8+T cells and NK cells. Murine IL-12 also promotes the development of proinflammatory/TH1-like CD4+ cells and cytotoxic CD8+T cells. Murine IL-12 is a disulfide linked heterodimeric protein with a molecular weight of 70 kDa comprised of disulfide-bonded 35 kDA (p35) and 40 kDa (p40) subunits.

Formulation : |
Sterile filtered and then the protein was lyophilized from 0.3 mg/ml in 1 x PBS pH 6.3. |
Stability : |
The lyophilized protein, is stable for a few weeks at room temperature, but best stored at -200C. Reconstituted murine IL-12 should be stored in working aliquots at -200C. |
Purity : |
Greater than 95% by SDS-PAGE. Endotoxin level
is less than 0.1 ng per µg (1EU/µg).
|
Reconstitution : |
We recommend a quick spin followed by reconstitution in 1 x PBS pH 6.3-6.5 to a concentration of 0.3 mg/ml. This solution can then be diluted into other aqueous buffers and stored at 40C for 1 week or -200C for future use. |
Biological Activity : |
A. Determined by the stimulation of
IFN-gamma production by murine splenocytes co-stimulated with IL-12.
The expected ED50 for this effect is < 0.1 ng/ml, corresponding
to a specific activity of >1 x107 units/mg.
B. Determined by the dose-dependent stimulation of the proliferation
of Immature B Lymphocyte murine 2D6 cells. The expected ED50 for
this effect is 0.5 - 1.0 ng/ml.
